Drug Interactions

There are significant differences in drug interactions among the thiazolidinediones. Pioglitazone is metabolized by the cytochrome P450 isoform CYP3A4 (10). Thus, safety and efficacy could possibly be affected when pioglitazone is co-administered with other drugs metabolized by this enzyme and hence, blood-glucose should be monitored more carefully in such patients (10,107). in vitro data demonstrate that rosiglitazone is predominantly metabolized by CYP2C8, and to a lesser extent, 2C9.
